Its population in July 2008 was estimated to be about 41082, It is West Virginia's 14th most populous county. The county encompasses an area of 512.19 mi˛ (1326.57 sq km ), of which, 505.79 square miles (1309.99 sq km) of it is land and 6.4 square miles (16.58 sq km) of it is water, making it the 15th largest by size in West Virginia. It is ranks seventh in the size of its inland waterways in West Virginia.

The population density was 84.8 people per square mile (32.74/km˛), it would be the 17th most densely populated county in the U.S. state of West Virginia. In the Wayne County, 23.4% of the population was under the age of 18, 8.7% were 18 to 24, 27.7% were 25 to 44, 25.3% were 45 to 64, and 14.9% who were 65 or older.
