Rowlesburg Town is a town located in Preston County, West Virginia, in the Southern Region (South Atlantic) of United States. As per the 2000 census, town has a population of 613 inhabitants. As of July 2008, the estimated population was 613, It is the 88th smallest by population in the U.S. state of West Virginia. The town comprises an area of 1.14 mi˛ (2.95 square kilometre), of which, 1.05 square miles (2.72 square kilometre) of it is land and 0.08 square miles (0.21 square kilometre) of it is water, giving it the 135th largest by total area among West Virginia.

According to 2000 Census Bureau, there were 613 people, 260 households, and 164 families living in the town. The population density was 582 people per square mile (224.71/km˛), it is the 72nd least populated town in terms of population density. There were 304 housing units at an average density of 289.5 per square mile (111.78/km˛). The average household size was 2.36 and the average family size was 2.98. In the town the population is diverse age-wise, with 21.4% under the age of 18, 8.3% from 18 to 24, 26.9% from 25 to 44, 23.8% from 45 to 64, and 19.6% who were 65 or older. For every 100 females there were 88.6 males. For every 100 females age 18 and over, there were 82.6 males. The median age is 41.5 years, the median age of male residents at 39 and the median age of female residents at 43.8. The racial or ethnic makeup of the town was 604 White, 1 Black or African American, American Indian, 2 Asian, Native Hawaiian, 6 from two or more races and Ohter races.
