Ponce De Leon Town is one of the town of Holmes County, Florida, located towards the Southern Region (South Atlantic) of United States. In 2000, the population was 457. As of July 2008, the population was estimated at 465, making it Florida's 72nd smallest. It has total area of 4.96 sq mi (12.85 kmē), of which, 4.95 square miles (12.82 kmē) of it is land and 0.01 square miles (0.03 kmē) of it is water, it is the 332nd largest by area in the U.S. state of Florida.

As of the census of 2000, there were 457 residents, 200 households, and 131 families in the town. The population density was 92.3 people per square mile (35.64/kmē), it is the 20th least densely populated town in the U.S. state of Florida. There were 233 housing units at an average density of 47.1 per square mile (18.19/kmē). It has an average household size was 2.29 persons, and the average family size was 2.82 persons.
