Perry County is a one of the county from Missouri's 115 counties and independent city, located in Midwestern Region (West North Central) of United States. As of 2000 the population was 18132. The county's estimated population as of July 2008 was 18743, making it the 56th most populous county in Missouri. The county comprises an area of 484.28 sq mi (1254.28 square kilometre), of which, 474.67 square miles (1229.39 square kilometre) of it is land and 9.61 square miles (24.89 square kilometre) of it is water, making it Missouri's 25th smallest county by total area.

The population density was 38.2 people per square mile (14.75/kmē), giving it the 47th most densely populated county among the Missouri. For every 100 males age 18 and over, there were 102.6 females.
