Mayes County, Oklahoma is a county located in the Southern Region (West South Central) of United States. The population of county in 2000 stood at 38369 as per the Census data. As of July 2008, the estimated population of the county is 39912, giving it the 25th largest population among Oklahoma's county. It covers total area of 683.51 mi˛ (1770.28 square kilometre), of which, 656.14 square miles (1699.39 square kilometre) of it is land and 27.37 square miles (70.89 square kilometre) of it is water, it is the 21st smallest in terms of geographical size.

In the county the population is diverse age-wise, with 26.6% under the age of 18, 8.6% from 18 to 24, 26.2% from 25 to 44, 23.8% from 45 to 64, and 14.9% who were 65 or older. For every 100 females there were 98.4 males. For every 100 females age 18 and over, there were 95.4 males. Its population was estimated at 39912 in July 2008, The racial makeup of the county was 71.11% White, 0.57% Black or African American, 20.01% American Indian, 0.41% Asian, 0.02% Native Hawaiian, and 7.87% from two or more races.
