Knox County is a one of the county from Missouri's 115 counties and independent city, located in Midwestern Region (West North Central) of United States. As of 2000 the population was 4361. According to July 2008 Census Bureau estimates, the population of the county is 4020, It is the third least populous county in the U.S. state of Missouri. It has total area of 506.77 mi˛ (1312.53 square kilometre), of which, 505.71 square miles (1309.78 square kilometre) of it is land and 1.07 square miles (2.77 square kilometre) of it is water, making it Missouri's 32nd smallest county by total area.

The population density was 8.6 people per square mile (3.32/km˛), it is Missouri's fourth least densely populated county. There were 2317 housing units at an average density of 4.6 per square mile (1.78/km˛). It has an average household size was 2.38 persons, and the average family size was 2.93 persons. In the county the population was spread out, with 24.9% under the age of 18, 6.2% from 18 to 24, 23.7% from 25 to 44, 23.9% from 45 to 64, and 21.2% who were 65 or older. For every 100 females there were 92.9 males. For every 100 females age 18 and over, there were 91.6 males. In July 2008, its estimated population was 4020, The racial makeup of the county was 3956 White, 5 Black or African American, 1 American Indian, 4 Asian, 0 Native Hawaiian, and 54 from two or more races.
