Jessamine County is a county located in Kentucky, in the Southern Region (East South Central) of United States. The population of the county was 39041 as per the 2000 census data. The county's estimated population as of July 2008 was 46716, giving it the 19th largest population among Kentucky's county. The county extends throughout a territory of 174.45 square miles (451.82 sq km ), of which, 173.13 square miles (448.4 sq km) of it is land and 1.32 square miles (3.42 sq km) of it is water, making it the eighth smallest by size in Kentucky.

The population density was 225.5 people per square mile (87.07/kmē), it is Kentucky's tenth most densely populated county. There were 14646 housing units at an average density of 84.6 per square mile (32.66/kmē). It has an average household size was 2.69 persons, and the average family size was 3.05 persons. In the county the population was spread out, with 26.4% under the age of 18, 11.6% from 18 to 24, 31.1% from 25 to 44, 21.4% from 45 to 64, and 9.5% who were 65 or older. For every 100 females there were 96.6 males. For every 100 females age 18 and over, there were 92.8 males. It has an estimated population (as of July 2008) of 46716, The racial makeup of the county was 94.23% White, 3.44% Black or African American, 0.19% American Indian, 1% Asian, 0.05% Native Hawaiian, and 1.1% from two or more races.
