Holmes County is a county located in Mississippi, in the Southern Region (East South Central) of United States. In 2000, county's population was 21609 inhabitants. Its population in July 2008 was estimated to be about 20595, giving it the 48th largest population among Mississippi's county. The county has a total area of 764.18 square miles (1979.22 kmē), of which, 756 square miles (1958.03 kmē) of it is land and 8.18 square miles (21.19 kmē) of it is water, making it the 12th largest by size in Mississippi.

The county's estimated population as of July 2008 was 20595, The racial makeup of the county was 3763 White, 16619 Black or African American, 31 American Indian, 42 Asian, 1 Native Hawaiian, and 139 from two or more races.
