Hamlin County is one of the 66 county of South Dakota, located towards the Midwestern Region (West North Central) of United States. As of 2000 the population was 5540. Its population in July 2008 was estimated to be about 5660, It is the 32nd largest in terms of population. It covers total area of 537.96 sq mi (1393.31 square kilometre), of which, 506.86 square miles (1312.76 square kilometre) of it is land and 31.1 square miles (80.55 square kilometre) of it is water, it is the tenth smallest by area in the U.S. state of South Dakota.

The population density was 10.9 people per square mile (4.21/kmē), making its South Dakota's 19th most densely populated county. There were 2626 housing units at an average density of 5.2 per square mile (2.01/kmē). The average household size was 2.62 people, and the average family size was 3.22 people. In the county Population was distributed with 29.4% under the age of 18, 6.9% from 18 to 24, 24% from 25 to 44, 20.5% from 45 to 64, and 19.2% who were 65 or older. For every 100 females there were 98.9 males. For every 100 females age 18 and over, there were 96.8 males. Its population was estimated at 5660 in July 2008, The racial makeup of the county was 98.22% White, 0.12% Black or African American, 0.76% American Indian, 0.18% Asian, 0% Native Hawaiian, and 0.72% from two or more races.
