De Soto City is a city located in the Jefferson County, Missouri, United States. In 2000, city's population was 6375 inhabitants. Its population in July 2008 was estimated to be about 6477, It is the 108th largest by population in the U.S. state of Missouri. The city spans an area of 3.81 sq mi (9.87 kmē), giving it the 160th largest by total area among Missouri.
