Brule County is a one of the county from South Dakota's 66 counties, located in Midwestern Region (West North Central) of United States. The county has a population of 5364 as per the 2000 census data. As of July 2008, the population was estimated at 5205, making it the 36th most populous county in South Dakota. It is spread over 846.49 mi˛ (2192.4 sq km), of which, 818.96 square miles (2121.1 sq km ) of it is land and 27.54 square miles (71.33 sq km) of it is water, making it South Dakota's 38th largest county by total area.

According to 2000 Census Bureau, there were 5364 residents, 1998 households, and 1328 families in the county. The population density was 6.5 people per square mile (2.51/km˛), giving it the 30th most densely populated county among the South Dakota. There were 2272 housing units at an average density of 2.8 per square mile (1.08/km˛). The average household size was 2.49 persons, and the average family size was 3.14 persons. In the Brule County, 30.5% of the population was under the age of 18, 6.8% were 18 to 24, 24.7% were 25 to 44, 21.2% were 45 to 64, and 16.9% who were 65 or older. For every 100 females there were 93.2 males. For every 100 females age 18 and over, there were 92.1 males. As of July 2008, the estimated population was 5205, The racial or ethnic makeup of the county was 4577 White, 14 Black or African American, 520 American Indian, 22 Asian, 1 Native Hawaiian, and 71 from two or more races.
